Improving Oral Dosage Formulations: Microencapsulation for Controlled and Immediate Release
The need for novel drug delivery technologies is on the rise as pharmaceutical companies face steep patent expiration cliffs and a dwindling number of candidates in the R&D pipeline. Patented drug delivery technologies can enable new formulations of existing drugs thereby extending product life-cycles and protecting from generic competition while simultaneously improving drug performance including release rate, taste-masking, and format flexibility. With the oral delivery market representing the largest drug delivery route, there is significant opportunity and need for new drug delivery technologies that can specifically address this market segment.
This presentation will focus on Orbis Biosciences’ patented Precision Particle Fabrication technology and its ability to develop novel oral formulations. We will discuss how the technology can enable precise dosage control of release profiles including immediate, extended, and delayed burst for both poorly and highly water-soluble APIs while enhancing taste-masking and mouth feel. We will show how these precision microparticle drug-intermediates can be integrated into a variety of formulations including solutions, tablets, sachets, and capsules. In addition, we will discuss the key differentiating factors of the Orbis technology as it compares to current manufacturing processes. Finally, we will discuss the ability of Precision Particle Fabrication to serve as a true platform technology capable of developing formulations for oral, injectable, and topical delivery routes.
Key takeaway points:
Benefits of Orbis' Precision Particle Fabrication
Format flexibility (suspensions, tables, capsules, sachets, etc.)
Release profiles: including immediate, extended and enteric
Compatible with both hydrophobic and hydrophilic APIs
High loading with minimal burst
Comparison of Precision Particle Fabrication melt-spray compared to spray drying and hot-melt extrusion
Particle size uniformity
Material flexibility
Single-step process (no grinding or sieving required)
Solvent free
Reduced batch-to-batch variability
Precision Particle Fabrication is broadly applicable
Controlled release injectable depots
